I find my black patent Miu Miu nappa charm super comfy and it holds a ton. My Chanel WOC is comfy too - it's so lightweight and the chain never digs. My LV Nimbus in anthracite is amazing too - so smooshy and fits perfectly on my shoulder.

My belenechandia Whisper to me is my most comfy bag because you can wear it messanger style, or over the shoulder where it stays without slipping or by the double handle in the croo of my arm. It's super light weight and superb craftmenship. It's my go-to bag for which I neglect all my other designer bags even BV, Chanel and Bal.
 
Kooba Devin. Softest strap ever. Bag opening hits right at the top of my waist so I can get into it while keeping it on my shoulder. I have it in the bluish/lavender color. Definetely the most comfortable bag I own.
 
My black Bally hobo shoulder bag with gold hardware is really comfortable - it's small so it's light and I can just toss in mini wallet, phone, keys, slip it onto my shoulder and I'm good to go.
 
I would have to say my Longchamp pliage tote. It's comfortable to carry even my 6lb laptop inside. Even more so than a backpack, but maybe I'm just a shoulder bag person.
 
My Kale Utility bag is so comfortable. The shoulder straps are flat. So when the bag is heavy, the shoulder straps don't feel as though they are digging into my shoulder, as rolled shoulder straps often do. This has become so important to me that I now only look for bags with flat shoulder straps, unless it is a smaller bag that I know I will never stuff until it is heavy.
 
I love my Mulberry A4 Roxanne tote. It's durable, chic and the shoulder straps can fit over my bulkiest winter coat. I can fill it up to the brim and it still looks good.
