Hopefullly I can offer a little glimmer of hope on this. See what LMB says, and hopefully they can help.

My cat peed on my husband's wheelie bag suitcase a couple of times a few years back when she was mad that he was gone so much. He left it lying on the floor flat, so she climbed on top and peed on it -- it was on the exterior and dripped inside, so throughout the bag pretty well. And she peed on it a couple of times, not just once. It was a $600+ suitcase, so figured I'd try to save it. I used Out! pet odor eliminator (this stuff - it's an enzyme cleaner like tiramisu recommends) and it totally got the smell out. Completely. No lingering hint of cat pee at all, and I have a super-picky nose. I had to apply it a couple of times, and I let it sit for a long time, but it was totally worth it.

The hitch is that you have to use the product with warm water, so your bag is probably going to dry stiff, and then you're going to have to work get it back to its nice squishiness. Maybe you can at least do this and then LMB can recondition it from there. I have to say, as a pet owner, that I don't think any of the powdered stuff is going to work, either the baking soda stuff (which I've used in the litterbox before) or the enzyme stuff, since you need to lift out what's in the leather fibers or it will smell again.

Good luck - I hope LMB has good news for you, but you may be able to get the smell out and then just need to soften it back up from being waterlogged...
